108 Deciphering the immune mechanism of autoreactive B cells in pemphigus

Pemphigus represents a group of severe autoimmune bullous diseases driven by the production autoantibodies autoreactive B cells that are mainly directed against desmosomal 1 and 3 adhesion proteins. A better understanding biology will allow us to develop new therapeutic strategies targeting specific depletion effector while maintaining immune surveillance. Our objective was perform proteomic transcriptomic analysis on Dsg3+ at single cell resolution using innovative analytic tools technologies. The characterization frequency phenotype subsets high dimensional coupled bulk RNA sequencing have shown presence subset in pemphigus patients associated with expression CD83 compared remission or healthy donors. use “index sorting” versus Dsg3- from donors has allowed identify unique activated pathways marker(s) including expressed these cells. Analysis lesional skin patient immunofluorescence revealed ectopic lymphocytic structures enriched CD83+ In addition, activation leads anti-Dsg3 vitro. results involved pemphigus.
